About This Section
| Entrez Gene summary for SSH1: The ADF (actin-depolymerizing factor)/cofilin family (see MIM 601442) is composed of stimulus-responsive mediators ofactin dynamics. ADF/cofilin proteins are inactivated by kinases such as LIM domain kinase-1 (LIMK1; MIM 601329). TheSSH family appears to play a role in actin dynamics by reactivating ADF/cofilin proteins in vivo (Niwa et al., 2002(PubMed 11832213)).(supplied by OMIM) UniProtKB/Swiss-Prot: SSH1_HUMAN, Q8WYL5Function: Protein phosphatase which regulates actin filament dynamics. Dephosphorylates and activates the actinbinding/depolymerizing factor cofilin, which subsequently binds to actin filaments and stimulates their disassembly.Inhibitory phosphorylation of cofilin is mediated by LIMK1, which may also be dephosphorylated and inactivated by thisprotein Gene Wiki entry for SSH1 |

About This Section
| UniProtKB/Swiss-Prot: SSH1_HUMAN, Q8WYL5 (See
protein sequence)Recommended Name: Protein phosphatase Slingshot homolog 1 Size: 1049 amino acids; 115511 Da
Subunit: Interacts with actin and this stimulates phosphatase activity. Also interacts with LIMK1 and with the 14-3-3proteins YWHAB, YWHAG, YWHAQ, and YWHAZ. Interaction with 14-3-3 proteins inhibits phosphatase activity and alsoblocks recruitment to lamellipodia and stimulation by actin
Subcellular location: Cytoplasm, cytoskeleton. Cell projection, lamellipodium. Cleavage furrow. Midbody. Note=Alsorecruited to actin rich membrane protrusions such as lamellipodia, which may allow local control of actin dynamics atsites of cell locomotion. Also localized to the cleavage furrow and the midbody during cytokinesis
Miscellaneous: Tyrosine phosphatase activity has not been demonstrated for this protein to date
Sequence caution: Sequence=BAA92536.1; Type=Erroneous initiation;
Secondary accessions: Q6P6C0 Q8N9A7 Q8WYL3 Q8WYL4 Q9P2P8Alternative splicing: 5 isoforms: Q8WYL5-1 Q8WYL5-2 Q8WYL5-3 Q8WYL5-4 Q8WYL5-5 Explore the universe of human proteins at neXtProt for SSH1: NX_Q8WYL5
Post-translational modifications:
Phosphorylated. Inhibitory phosphorylation by PAK4 promotes binding to YWHAZ. Phosphorylation at Ser-978 is decreasedby stimuli which promote actin reorganization and lamellipodia formation. Can be dephosphorylated and activated byPPP3CA/calcineurin A. Phosphorylation decreases immediately prior to telophase1
